Plants exhibit selectivity in ion uptake by roots. Even for ions at the same concentration in the environment, their absorption rates vary. For instance, in ammonium sulfate solution, plant roots absorb ammonium ions faster than sulfate ions; while in sodium nitrate solution, roots take up nitrate ions more rapidly than sodium ions.
Salts whose cations are absorbed more than anions, resulting in a drop in the pH of the medium, are defined as physiologically‑acidic salts. Salts whose anions are absorbed more than cations, leading to an increase in the pH of the medium, are defined as physiologically‑alkaline salts.
Plant Physiologically‑Acidic Salt Solution (Aladdin Cat.No.: P1526083) refers to salts where cations are absorbed in greater quantities than anions, which causes the pH of the medium to decrease.Plant Physiologically‑Alkaline Salt Solution (Aladdin Cat.No.: P1526084) refers to salts where anions are absorbed in greater quantities than cations, which causes the pH of the medium to increase. The plant physiological control salt solution is mainly composed of potassium dihydrogen phosphate, magnesium sulfate, calcium chloride, etc., with a pH of approximately 5.8. The mass concentration of potassium dihydrogen phosphate is 0.0125 %, and magnesium sulfate is 0.0125 %. It does not contain ammonium sulfate or sodium nitrate. This reagent is for scientific‑research use only and is not intended for clinical diagnosis or other applications.
Operating Procedures (For Reference Only)
1. Select robust plant seedlings of uniform size. Remove the endosperm with a razor blade to obtain experimental materials.
2. Cover the bottle mouth with black plastic film and fasten it with rubber bands. Punch several small holes in the film using tweezers. Insert the seedling roots through the holes into the solution, and incubate at ambient temperature.
3. After three days, take out the seedlings. Top up the solution with distilled water to the marked level, mix thoroughly, and measure its pH value.
4. Measure and calculate the experimental results according to the table below:
|
Calculation:
The relative uptake of anions and cations in solution by plant seedlings can be analyzed by comparing the ΔpH of the experimental group with that of the control group.
Precautions
1. Adjust and measure the pH value of the solution accurately.
2. Select plant seedlings with consistent growth status and avoid damaging their root systems.
3. Use the reagent as soon as possible after opening to prevent adverse effects on subsequent experimental results.
4. For your safety and health, wear a laboratory coat and disposable gloves during operation.
