Natural pectin substances exist widely in the fruits, roots, stems, and leaves of plants in the forms of protopectin, pectin, and pectic acid. They are components of the cell wall, accompanying cellulose, and constitute the intercellular adhesive layer between adjacent cells, firmly bonding plant tissue cells together. Protopectin is a water-insoluble substance, but under the action of chemical reagents such as acids, alkalis, or salts, as well as enzymes, it can undergo hydrolysis and transform into water-soluble pectin. Pectin, also known as polygalacturonic acid, is a linear polymer composed of D-galacturonic acid units linked by α-1,4 glycosidic bonds. It is essentially a linear polysaccharide polymer containing several hundred to about 1000 anhydrogalacturonic acid residues, with a corresponding average relative molecular mass ranging from 50,000 to 150,000. Protopectin is a water-insoluble substance. In immature fruits or plant tissues, pectin substances are mostly combined with cellulose and exist in the form of protopectin, which makes the fruits or other plant tissues hard and crisp. The detection principle of this kit is as follows: Pectin substances are hydrolyzed to generate galacturonic acid, which then undergoes a condensation reaction with carbazole in a sulfuric acid solution to form a purplish-red compound. The color intensity of this compound is proportional to the concentration of galacturonic acid. The color of the compound reaches its maximum depth within 1-2 hours of the reaction. When the reaction solution exhibits the deepest color, the absorbance is measured at a wavelength of 530 nm. By comparing with a standard curve, the content of protopectin or soluble pectin in the sample is calculated. This kit is primarily used for the quantitative determination of protopectin content in plant tissues or fruits, and can also be used for the quantitative determination of soluble pectin content in plant tissues or fruits, thereby allowing the calculation of total pectin content (which is the sum of protopectin content and soluble pectin content). This product is for research use only and is not intended for use in diagnostic procedures.
