The purpose of fixation is to preserve the original morphological structure of cells and tissues. Fixatives can prevent endogenous lysosomal enzymes from autolyzing tissues and cells, and inhibit the growth of bacteria and molds. Fixatives are mainly classified into aldehyde fixatives, mercury fixatives, alcohol fixatives, oxidant fixatives, picrate fixatives, etc. The most commonly used are formalin (aldehyde) and ethanol (alcohol).
Regaud's fixative is composed of dichromate solution and formalin. It is a special fixative for bone tissue, and is rarely used for other tissues. In some cases, it cannot be used for bone tissue staining that contains mercury.
This reagent is only for research use, not suitable for clinical diagnosis or other purposes.

Procedure (for reference only):
Operate according to the specific experimental requirements. Immediately before use, mix Regaud solution and formalin solution thoroughly at a ratio of 1:1, and use promptly.
Precautions:
1. The fixation time varies with the thickness of tissue sections. Proper selection of tissue samples facilitates penetration of fixative. The suitable thickness for routine biopsy tissues is 2–4 mm, generally not exceeding 6 mm.
2. The volume of fixative should be sufficient. Generally, the volume ratio of fixative to tissue block should be greater than 10:1.
3. Temperature has a significant effect on fixation. Raising the temperature can accelerate fixation, but the temperature should not be too high.
4. Fresh tissues should be fixed promptly after removal. If immediate fixation is not possible, they should be stored in physiological saline and sent for examination promptly.
5. Please use the reagent as soon as possible after opening to prevent affecting subsequent experimental results.
