Panoramica

Resistant starch (RS) refers to starch that cannot be digested by enzymes in the human small intestine and is partially or completely fermented in the large intestine. RS is a component of total dietary fiber. Determination method for resistant starch: The sample is incubated with pancreatic α‑amylase and amyloglucosidase (AMG) in a shaking water bath at 37 °C for 16 hours. During this period, non‑resistant starch is hydrolyzed to D‑glucose by the combined action of the two enzymes. After incubation, an equal volume of ethanol or industrial methylated spirit (IMS, denatured ethanol) is added to terminate the reaction. The solution is centrifuged, and the supernatant is collected and retained. The residue at the bottom (a flocculent pellet) is the RS in the sample. The pellet is washed with aqueous IMS or 50 % (v/v) ethanol, centrifuged, and the washing/centrifugation step is repeated once. The supernatants obtained from the washing steps are combined with the previously collected supernatant. Any remaining liquid in the tube is carefully decanted, and the pellet is placed in an ice‑water bath and dissolved in 2 M KOH with vigorous stirring using a magnetic stirrer. The solution is then neutralized with acetate buffer, and the starch is quantitatively hydrolyzed to glucose by AMG. D‑Glucose is measured using glucose oxidase/peroxidase reagent (GOPOD), which gives the RS content in the sample. Non‑resistant starch (soluble starch) can be determined by pooling the combined supernatants, making up to 100 mL, and measuring D‑glucose with GOPOD.
