Sabouraud Dextrose Broth (SDB) is a common liquid medium for fungi. It is weakly acidic and rich in glucose and organic nitrogen sources, supporting the growth of yeasts and moulds while inhibiting most bacteria. It is routinely used for fungal culture in pharmaceuticals and biological products, with an incubation temperature of 20–25 °C.
Principle and Purpose:
A mixture of peptic digest of animal tissue and pancreatic digest of casein supplies nitrogen sources, vitamins and growth factors; glucose serves as the carbon source.
Preparation Method:
Weigh 30.0 g of this product and add to 1 L of purified water. Heat and stir until completely dissolved, dispense into suitable containers, sterilize by autoclaving at 121 °C for 15 min, and set aside for use.
Scope of Application:
| Application | Category |
| Non-selective enrichment | Moulds and yeasts |
Formula (g/L):
| Equal mixture of peptic digest of animal tissue and pancreatic digest of casein | 10.0g |
| Glucose | 20.0g |
| Final pH (25 °C) | 5.6±0.2 |
Quality Control:

Testing shall be carried out in accordance with the Chinese Pharmacopoeia using Candida albicans and Aspergillus niger.
| QC Strain and Strain No. | Growth Performance / Characteristics |
| Candida albicans CMCC(F)98001 | Good growth, medium turbidity observed |
| Aspergillus niger CMCC(F)98003 | Good growth; white mycelia on liquid surface with a small amount of black spores |
Inoculation level: 10–100 CFU
Incubation conditions: 20–25 °C for 48–72 hours
Reference Standard:
Chinese Pharmacopoeia
Precautions:
1. Take necessary protective measures when weighing the medium to avoid physical discomfort.
2. The medium shall be stored in a dark and dry place. Tighten the bottle cap immediately after use to prevent moisture absorption and caking.
3. The used medium shall be autoclaved at 121℃ for 30 minutes before disposal.
4. This product is not intended for clinical testing.
