The purpose of fixation is to preserve the original morphology and structure of cells and tissues. Fixatives prevent autolysis of tissues and cells by endogenous lysosomal enzymes, inhibit the growth of bacteria and molds, and inactivate enzymes by altering protein internal structures through coagulation, formation of additive compounds, and other mechanisms. Fixatives also induce physical changes in cellular and extracellular components. Fixatives are generally classified into aldehyde-based, mercury-based, alcohol-based, oxidant-based, and picrate-based types. The most commonly used are formaldehyde (from the aldehyde group) and ethanol (from the alcohol group). Glutaraldehyde fixatives can cause deformation of the protein α-helix structure, which is unfavorable for peroxidase staining. Glutaraldehyde fixatives exhibit rapid fixation but poor penetration.
This product is a buffered composite fixative, primarily formulated with acetic acid, sodium acetate, formaldehyde, and purified water in scientifically determined proportions. It features a weak acid buffer system with stable pH, balancing fixation penetration and sample morphology preservation. With a mature formulation and good reproducibility, it is a classic fixative specifically designed for parasitological testing. It is primarily suitable for rapid fixation, long-term preservation, sample concentration, and subsequent permanent staining and slide preparation of various intestinal parasite samples. It effectively preserves the structures of parasite eggs, larvae, protozoa, and other parasitic forms, preventing autolysis, deformation, disintegration, and detachment, while maximizing the retention of intact morphology and fine structural details. It is applicable to fecal parasite examinations, parasite specimen preparation, teaching and research specimen storage, and routine laboratory staining and identification.
This reagent is intended for research use only and is not suitable for clinical diagnosis or other applications.
Operating Instructions (For Reference Only):
1.Sample Preparation: Collect fresh fecal samples or isolated parasite specimens promptly. Remove large debris as much as possible and minimize the time of sample exposure ex vivo to prevent premature autolysis of parasites.
2.Fixation Immersion: Add the sample to be processed into the SAF fixative. The volume of fixative should be 10–15 times the sample volume. Mix thoroughly to ensure that all parasite eggs and organisms are completely submerged in the fixative.
3.Fixation Time: Routine parasite samples can be adequately fixed at room temperature within 10–30 minutes. For long-term preservation, samples may be directly left immersed in the fixative without the need for fluid replacement.
4.Subsequent Procedures: After fixation, samples can be directly subjected to concentration, sedimentation, smear preparation, or permanent staining (e.g., hematoxylin and eosin staining) and microscopic examination. Alternatively, they may be sealed and stored for long-term use.

2.Ensure that samples are completely submerged during fixation to prevent localized exposure, which may lead to parasite autolysis and damage. Process different samples separately to avoid cross-contamination.
3.This product is intended for laboratory parasite sample fixation and research testing only. It is strictly prohibited for use on humans, living tissues, or clinical treatment.
