Plant tissue‑culture, also known as axenic culture or in‑vitro culture, is a technique based on the theory of plant cell totipotency. Under suitable artificial medium and environmental conditions, isolated plant organs (such as roots, stems, leaves, flowers, shoot‑tips, fruits, etc.), tissues (such as cambium, epidermis, surface‑layer cells, pith cells, endosperm, etc.), cells (megaspores, microspores, somatic cells, etc.) and protoplasts can be induced to produce callus, adventitious buds and adventitious roots, and eventually develop into intact plantlets. Commonly‑used media for plant tissue‑culture include MS, NT, B5, N6, White, HDKW, Litvay, WPM and so on.
SH medium (Schenk & Hildebrandt medium) was designed by Schenk and Hildebrandt in 1972. Similar to B5 medium, it is a medium with a high concentration of inorganic salts and is mainly used for suspension‑cell culture of dicotyledonous and monocotyledonous plants. Compared with the basic components of MS, B5, N6, White, H and other media, SH medium uses disodium hydrogen phosphate instead of potassium dihydrogen phosphate, ammonium dihydrogen phosphate in place of ammonium nitrate or ammonium sulfate, and contains a relatively‑high‑content of myo‑inositol, which is its prominent feature. Thanks to its high‑ion‑concentration formula, slight deviations in some components during preparation, storage and sterilization will not affect the ionic equilibrium.
SH Vitamin Solution (100×) is a vitamin mixture widely used in plant tissue‑culture, mainly composed of myo‑inositol, nicotinic acid, pyridoxine hydrochloride, thiamine and other ingredients. It can be used as an additive for the basal salts of corresponding plant‑tissue‑culture media. This product is a 100 × stock solution. Add 10 mL of this solution to every 1 L of culture medium, and it will not change the pH value of the medium. This product is non‑sterile. It is recommended to perform sterilization treatment after preparing the working‑solution to prevent microbial contamination. This reagent is for research‑use‑only and shall not be applied for clinical‑diagnosis or other purposes.
Operating Procedure (For Reference Only):
1. Directions for use: Add 10 mL of this product into 1 L of medium solution and mix well. Adjust to the required pH value, generally 5.8±0.2.
2. If necessary, autoclave at 121 °C for 15‑20 min for plant‑tissue‑culture applications. Additional carbon‑sources, gelling agents, plant‑hormones or plant‑growth‑regulators may also be supplemented.
Precautions:
1. Select an appropriate culture‑medium according to your experimental materials and plant species. Optimal culture conditions are best determined experimentally.
2. Use the reagent as soon as possible after opening to avoid adverse impacts on subsequent experiments.
3. For your safety and health, please wear a lab coat and disposable gloves during operation.
