Xylanase (EC 3.2.1.8) is mainly produced by microorganisms and catalyzes the hydrolysis of xylan. It is also known as pentosanase or hemicellulase. This enzyme degrades cell wall components and β‑glucan in raw materials used in the brewing and feed industries, reducing mash viscosity, promoting the release of active substances, and reducing non‑starch polysaccharides in feed to enhance nutrient absorption. It is therefore widely used in both industries. Acid xylanase (ACX) is typically isolated from acid‑tolerant fungi, bacteria, and certain molds. Assay principle: ACX hydrolyzes xylan into reducing oligosaccharides and monosaccharides under acidic conditions. These reducing sugars react with 3,5‑dinitrosalicylic acid (DNS) in a boiling water bath, producing a colored product with a characteristic absorption peak at 540 nm. The color intensity is proportional to the amount of reducing sugars generated. ACX activity is calculated by measuring the rate of increase in absorbance at 540 nm.
