Overview

Product Description
STM Solution (2M, pH 7.4), also known as Sucrose-Tris-MgCl₂ Solution, is mainly composed of 2 M sucrose, 10 mM Tris and MgCl₂, with a pH value of approximately 7.4. It is sterilized by filtration and primarily used for the isolation of root tips or epidermis from plant tissues. This reagent is for research use only and not intended for clinical diagnosis or any other purposes.

Materials to Be Prepared by Users
Mortar and pestle, centrifuge, gauze

Operating Procedures (For Reference Only)
1. Select the anterior 1-2 mm segment of root tips, weigh about 4 g of the sample, rinse it thoroughly, cut it into small pieces and place the pieces into a mortar. Add 20 ml of pre-chilled STM Solution (2M, pH 7.4) at 4 °C, and grind the mixture into homogenate.
2. Filter the homogenate through gauze, collect the filtrate, and centrifuge it at 500 × g for 30 seconds at 4°C.
3. Discard the supernatant, and resuspend the pellet in a small volume of STM Solution (2M, pH 7.4). The resulting suspension contains root tip meristematic cells. If the pellet contains a large amount of cell debris after the first centrifugation, resuspend the cells and centrifuge again at 500 × g for 30 seconds at 4°C.

Precautions
1. Ensure strict aseptic technique during operation.
2. Maintain the temperature at 4°C as much as possible throughout the entire process.
3. Use the reagent as soon as possible after opening to avoid affecting subsequent experimental results.
4. For your safety and health, wear a lab coat and disposable gloves during operation.
