This product is a high‑concentration aqueous sucrose solution formulated to an accurate molar concentration of 2.5 M.
For density‑gradient centrifugation applications, it serves as a critical supporting reagent for the isolation of subcellular particles and viruses using cell separation medium. Mix the stock cell‑separation medium with this product at a 9:1 volume ratio to prepare an isotonic stock master solution. Further dilute the master solution with 0.25 M sucrose solution to the target density for isopycnic density‑gradient centrifugation and separation of organelles including mitochondria, lysosomes and microsomes, as well as viruses and exosomes. The sucrose‑based system effectively prevents aggregation of subcellular components under salt‑ion conditions and preserves the structural and functional integrity of particles.
In addition, this product can be used as a cryoprotectant or osmotic buffer component to maintain the structural integrity of biological samples under low‑temperature conditions. It is a fundamental reagent for laboratory biochemical experiments and tissue culture.
Instructions for Use
1.Sterile handling: Inspect the bottle seal for integrity before use. Aspirate the required volume with a pipette or sterile graduated cylinder inside a biosafety cabinet. Avoid repeated freeze‑thaw cycles and repeated cross‑contaminating withdrawals.
2.Preparation of isotonic stock master solution (SIM): Mix the stock cell‑separation medium with this product at a 9:1 volume ratio (e.g., 90 mL stock cell‑separation medium + 10 mL 2.5 M sucrose solution). Slowly add this product into the stock cell‑separation medium, and mix thoroughly using a magnetic stirrer or vortex mixer.
3.Dilution to target working solution: Dilute the isotonic stock master solution with 0.25 M sucrose solution to the target working‑solution density according to experimental requirements. Prepare diluted working solutions fresh before use to prevent contamination or density drift upon prolonged storage. Refer to the cell‑separation medium manual for detailed volume‑calculation formulas.
4.In‑situ gradient separation: Directly mix the sample to be separated (subcellular particles or viruses) with the diluted working solution, then perform high‑speed centrifugation. A continuous density gradient forms spontaneously during centrifugation, and target particles migrate to their isopycnic positions.
5.Other applications: When used for osmotic adjustment or cryoprotection, dilute to the target concentration with sterile deionized water, calibrate osmotic pressure, perform strict aseptic manipulation, and prepare fresh for immediate use.
Precautions
1.Tighten the cap after opening to avoid concentration shift caused by water evaporation and microbial contamination.
2.If crystals precipitate after long‑term storage (especially under low‑temperature conditions), warm the bottle gently in lukewarm water and shake thoroughly until crystals are fully dissolved prior to use. Discontinue use immediately if turbidity or visible particulate matter is observed.
3.This product is intended for the isolation of subcellular particles and viruses. For cell samples such as lymphocytes and monocytes, use NaCl‑based systems (1.5 M NaCl Solution).
