Panoramica

  Nucleoside drugs are an important class of medicines used in modern clinical practice for the treatment of viral infections, malignant tumors, AIDS and other diseases. Currently, nearly 50% of antiviral drugs in use are nucleoside drugs. Thymidine phosphorylase (TP or TYMP), also known as thymidine orthophosphate deoxy-D-ribosyltransferase, is widely present in prokaryotic and eukaryotic cells. It is a key enzyme in the pyrimidine nucleoside salvage pathway in organisms and also an important enzyme for the enzymatic synthesis of nucleoside substances in vitro. 

  At present, three functions of this enzyme have been confirmed.The first function is as TYMP, with main roles including:① Participating in the pyrimidine salvage synthesis pathway, converting thymidine nucleotides into thymine and 2-deoxyribose-D-ribose-1-phosphate through dephosphorylation, thereby maintaining the dynamic balance of thymidine levels in the human body;② Participating in the metabolic process of the pyrimidine antimetabolite drug 5-fluorouracil in the human body, catalyzing the reaction between 5-fluorouracil and 2-deoxyribose 1-phosphate to produce 5-fluoro-2'-deoxyuridine, blocking the formation of thymidine, killing cells in the S phase of the proliferation cycle, and thus exerting anticancer activity. The second function is as platelet-derived endothelial cell growth factor (PD-ECGF). Studies have confirmed that this growth factor can stimulate endothelial cell chemotaxis and growth in vitro and promote angiogenesis in vivo. The third function is as gliostatin. It has been verified that TYMP and PD-ECGF are the same substance. Current research on TYMP mainly focuses on the evaluation of tumor therapeutic efficacy and prognosis.
