Paraformaldehyde (PFA) is one of the most commonly used fixatives in immunohistochemistry (IHC) and immunocytochemistry (ICC) research. Low-polymerization-degree paraformaldehyde, used as a substitute for conventional industrial formaldehyde aqueous solutions, offers significant advantages in the synthesis of pesticides, synthetic resins, coatings, and the production of fumigation disinfectants, as well as various other downstream formaldehyde products. It not only reduces energy consumption during dehydration but also greatly minimizes wastewater treatment volume, representing an environmentally friendly and socially beneficial green engineering solution.
Low-polymerization-degree paraformaldehyde, due to its higher active ingredient content compared to industrial formaldehyde and its solid particulate form, is advantageous for chemical synthesis, pharmaceutical manufacturing, and other industrial applications, particularly in syntheses requiring anhydrous formaldehyde as a raw material, where it finds extensive use.
The Tissue-cell Fixative (1% PFA) produced by Aladdin is primarily composed of paraformaldehyde, phosphate salts, and deionized water, with a pH of 7.4. This fixative is relatively mild, effectively preserving tissue antigenicity and fine structure. It is suitable for long-term preservation of tissue specimens and cell slides, making it highly appropriate for light microscopic immunochemical studies of tissues and cells.
This product is intended for research use only and shall not be used for other purposes.
Operating Procedure (For Reference Only):
Generally, tissue fixation time should be controlled at 4–12 hours. Larger specimens should have the fixation time appropriately extended. For cultured cells or cell culture slides, the fixation time should be controlled accordingly, except under special circumstances.
Precautions:
1.Paraformaldehyde solution (1% PFA) is somewhat irritating and corrosive. Once opened, the fixative effectiveness may decrease with prolonged storage.
2.Avoid excessively prolonging the fixation time, as this may cause excessive cross-linking of biological macromolecules. Fixation time varies depending on the thickness of the tissue samples.
3.The volume of fixative should be sufficient; generally, the volume ratio of fixative to tissue block should be greater than 10:1.
4.Temperature significantly affects fixation. Increasing the temperature can accelerate the fixation process, but the temperature should not be too high.
5.Freshly excised tissues should be fixed promptly. If immediate fixation is not possible, they should be stored in physiological saline and submitted for testing as soon as possible.
6.For your safety and health, please wear a lab coat and disposable gloves during operation.
