Cholesteryl esters are hydrolyzed to free cholesterol by cholesterol esterase. The free cholesterol is then oxidized by cholesterol oxidase to produce 4‑cholesten‑3‑one and hydrogen peroxide. In the presence of 4‑aminoantipyrine and phenol, the hydrogen peroxide is catalyzed by peroxidase to form a red quinone compound, which has an absorption peak at 510 nm. The color intensity is directly proportional to the free cholesterol content. The lower detection limit of this kit is 0.078 mmol/L, and it exhibits a good linear relationship within the range of 0.078 - 10 mmol/L.
