Zonisamide
Zonisamide（Aladdin Z163001 ≥98%；Z276568 ≥99%；Z408620 10mM in DMSO ） is an anticonvulsant medication used to manage partial-onset seizures, generalized tonic–clonic seizures, myoclonic seizures, and mixed seizure types in Lennox–Gastaut syndrome. Its mechanisms of action include inhibition of voltage-gated sodium channels, blockade of T-type calcium channels, and suppression of carbonic anhydrase activity. Zonisamide also reduces the development of neuronal hypersynchronization, a neural activity pattern associated with epileptiform seizures. Additionally, it modulates both GABAergic and glutamatergic neurotransmission.
Beyond epilepsy treatment, zonisamide has demonstrated activity in several other research contexts. In animal models of Parkinson’s disease, it decreases dopaminergic neuron loss in the nigrostriatal pathway via brain-derived neurotrophic factor (BDNF) signaling, improving both dopaminergic cell survival and motor performance.
In neuropathic pain models induced by chronic constrictive injury, zonisamide increases paw withdrawal latency, indicating analgesic and antinociceptive potential. These effects are observed across chemical, mechanical, and thermal hyperalgesia and allodynia models.
Zonisamide is also being studied for its potential anti-obesity effects. In clinical trials, it has been shown to aid weight reduction. In animal studies, zonisamide prevents weight gain, excessive eating, and elevated blood glucose levels when co-administered with second-generation antipsychotics such as olanzapine, which are commonly associated with metabolic side effects.
