Detection of activated platelets by flow cytometry

Operation method

Staining of activated platelets for double labeling detection by whole blood method

Materials and Instruments

Whole blood
Citrate or hirudin CD62p-FlTC CD42b-PE PBS

Move

1. Collect blood citrate or hirudin anticoagulation.

2. 10 ul CD62p-FlTC plus 10 ul CD42b-PE, plus 5 ul whole blood (sample tube); 10 ul IgG1-FITC plus 10 ul IgG1-PE, plus 5 ul whole blood (control tube); mix gently and incubate at room temperature for 15 min.

3. Add 2~3 ml PBS (1% PFA) Terminate the reaction and analyze the platelets by SSC-LOG/CD42b-PE circling.
