Overview

Catalase (CAT, EC 1.11.1.6) is widely present in animals, plants, microorganisms, and cultured cells. Its activity is closely associated with metabolic intensity, cold resistance, and disease resistance in organisms. Direct measurement of hydrogen peroxide (H₂O₂) has two major limitations: first, H₂O₂ itself is chemically unstable, which can lead to fluctuations in absorbance readings; second, components such as proteins in the sample also absorb light in the 240 nm UV region, severely interfering with result accuracy. To address these issues, this kit provides a simple, sensitive, and rapid assay. The principle is as follows: CAT catalyzes the decomposition of H₂O₂ to water and oxygen. The residual H₂O₂ in the reaction system reacts with a novel chromogenic probe to form a stable colored product with a maximum absorbance peak at 510 nm. By measuring the amount of this product (which reflects the decrease in H₂O₂), the CAT enzyme activity in the sample can be precisely calculated. This kit is suitable for the determination of CAT activity in a variety of biological samples, including plant tissues, serum, and plasma. Its key advantage is the shift of the detection wavelength from the UV region (240 nm) to the visible region (510 nm), eliminating the need for quartz cuvettes or UV-compatible microplates, and making it compatible with standard visible-light spectrophotometers.
