O precursor codificado é clivado para formar uma proteína ativa que contém uma cadeia pesada de 784 aminoácidos seguida de uma cadeia leve de 235 aminoácidos que estão ligadas por uma ou mais ligações dissulfureto. O ARNm é expresso apenas no intestino delgado e a proteína é encontrada nos enterócitos do duodeno e do jejuno proximal. O gene da enterocinase humana está localizado no cromossoma 21q21.
