Newly synthesized IL-18 is a precursor molecule to the signal peptide, which is enzymatically cleaved in order to activate it. In this ELISA method, in order to detect only mature (activated) IL-18, the coated antibody must be specific for activated IL-18. Some cells (e.g., human P B M C ) can also secrete IL-18 precursors under normal conditions, and culture supernatants from these cells can be used to detect one or both forms of IL-18. mixtures of anti-human or mouse IL-18 monoclonal antibodies can be used to detect the IL-18 precursor and/or mature form. In this protocol, the antibody to human IL-18 is specific for activated IL-18 only, and the anti-mouse IL-18 antibody detects both forms of mouse IL-18.
