Overview

This kit employs an alkaline buffer at pH 9.6–9.7 to extract nitrate ions from samples. During the extraction process, activated carbon is added to remove pigments, and a precipitant is added to eliminate proteins and other interfering substances. Taking advantage of the property that nitrate and nitrite ions exhibit equal absorbance at 219 nm in the ultraviolet region, the absorbance of the extract is measured. The result obtained is the combined absorbance of nitrate and nitrite. Since the nitrite content in fresh vegetables and fruits is extremely low and negligible, this measured value can be regarded as the absorbance attributable to nitrate. The nitrate content in the sample is then calculated based on a standard curve.
