Descripción general
Aconitase (ACO) is an enzyme in the tricarboxylic acid cycle that catalyzes the conversion of citrate to isocitrate. Citrate itself is not easily oxidized; under the action of aconitase, through dehydration and hydration reactions, the hydroxyl group is transferred from the β-carbon atom to the α-carbon atom, generating isocitrate which is readily dehydrogenated and oxidized, preparing for further oxidative decarboxylation reactions. This product provides a simple, convenient, and rapid method for assaying ACO activity, suitable for detecting samples such as animal tissues, plant tissues, cells, and serum (plasma). Its principle is that ACO catalyzes the conversion of citrate to isocitrate. Isocitrate is then oxidatively decarboxylated, reducing NAD⁺ to generate NADH, resulting in an increase in absorbance at 340 nm.
