In solid tissues and cells, cholesterol esters are not only components of the cell plasma membrane but also major constituents of intracellular lipid droplets. Excessive intracellular cholesterol accumulation is closely associated with the formation of atherosclerotic foam cells. Cholesterol determination in solid tissues and cells is far more complex than that in blood. This kit eliminates the need for toxic organic solvent extraction, cumbersome nitrogen drying, and lipid reconstitution steps. It employs high‑efficiency reagents for cell lysis and cholesterol extraction, with optimized enzymatic reactions and procedures, making it simple and easy to use while offering high sensitivity and a linear range of 20–5000 µmol/L. This kit utilizes the enzymatic method recommended by the WHO and China’s National Clinical Laboratory Operating Procedures, combined with the classic GPO Trinder enzymatic reaction. The principle is as follows: (1) Cholesterol esterase hydrolyzes cholesterol esters into free cholesterol; (2) Cholesterol oxidase oxidizes free cholesterol, producing hydrogen peroxide during the reaction; (3) A chromogenic reaction is catalyzed by peroxidase, producing a product with a maximum absorption peak at 550 nm, where the optical density is proportional to the cholesterol concentration; (4) The total cholesterol concentration and free cholesterol concentration are calculated separately, and the difference between the two values represents the cholesterol ester concentration.
