Diff-Quik staining is a rapid staining method developed and improved on the basis of Wright staining. It is commonly used for staining in cytological examinations. The staining solution is formulated according to the rapid staining method recommended by the World Health Organization (WHO). Similar to Wright Stain, it is modified based on the technical principle of Romanowsky Stain, and its staining results are also highly similar to those of Wright staining solution. However, Diff-Quik rapid staining requires a very short time, and staining can generally be completed within 90 seconds.
Diff-Quik Stain does not contain fixative solution. It is mainly used for staining blood smears, bone marrow smears, vaginal secretion smears, exfoliative cell smears, etc. It is very suitable for batch dip staining, with a clear background and no sediment. This staining solution is for research use only and is not suitable for clinical diagnosis or other purposes.

Protocol (For Reference Only):
1. Prepare blood smears or bone marrow smears using routine methods. Allow to air-dry or briefly pass over an alcohol lamp flame, then fix with methanol for 20–30 seconds.
2. Stain in Diff‑Quik Stain Solution I for 5–10 seconds.
3. Stain in Diff‑Quik Stain Solution II for 5–10 seconds.
4. Rinse with water and examine immediately under a microscope while wet.
5. Clear valuable slides with xylene, mount and preserve with neutral balsam.
Staining Results:
Nuclei, leukocytes: Dark blue
Matrix, lymphocytes: Purple
Cytoplasm, erythrocytes: Pink
Precautions:
1. Blood or bone marrow smears should be of uniform thickness to avoid affecting staining results.
2. Staining of blood cell smears requires fresh whole blood or EDTA anticoagulated blood.
3. For bone marrow smears, shake or fan rapidly in air after preparation to prevent cell shrinkage, deformation or hemolysis due to high humidity. Do not dry smears with high heat or open flame.
4. For vaginal discharge smears, fix fresh specimens promptly by flame or alcohol after smearing to prevent cell deformation.
5. If staining is too intense or too faint, adjust staining time or working solution concentration accordingly.
6. pH affects staining; slides must be clean and free of acid or alkali contamination.
7. For your safety and health, wear a lab coat and disposable gloves during operation.
