Antimicrobial Drug Test Tube Experiment
Antimicrobial Drug Test Tube Experiment
Antimicrobial drug test tube experiments can be used to (1) test bacterial susceptibility to antimicrobial drugs, (2) screen for the most efficacious drugs, and (3) control epidemics of bacterial infectious diseases.
Operation method
Antimicrobial Drug Test Tube Experiment
Principle
Concentration series dilution method when the drug is diluted into a series of different concentrations, mixed into the culture medium, adding a certain amount of test bacteria, the results are observed after incubation at a suitable temperature to find the minimum inhibitory concentration (MIC) of the drug.
Materials and Instruments
Staphylococcus aureus Move 1. Add 1 ml of broth medium to each tube according to aseptic procedures. Caveat 1. the accuracy of dilution: each dilution of a concentration of a pipette, more accurate than a pipette dilution in the end. 2. the concentration of the bacterial solution added: when the concentration of the bacterial solution is large, the minimum inhibitory concentration is high. 2. the concentration of the added bacterial solution: when the concentration of the bacterial solution is large, the minimum inhibitory concentration will be high, and vice versa. 3. bacterial age: it is generally believed that young bacteria are more sensitive, so the 6-hour culture of bacteria is used. 2. Add 1 ml of the original concentration of the drug to tube 1, mix well and then add 1 ml to tube 2, mix well and then add 1 ml to tube 3, and so on, and then remove 1 ml of the drug and discard it after double dilution to tube 9. The 10th tube was used as control without adding drug.
3. Add 0.1 ml of the above bacterial solution to each of tubes 1 to 10 and mix well.
4. Place in 37 ℃ constant temperature box after 18 ~ 24 hours of culture to observe the growth of bacteria in each tube.
5. Judgment of results: the highest dilution of the drug that completely inhibits the growth of bacteria is the lowest inhibitory concentration (MIC) of the drug on the bacteria.
