单 元 4.1)，生物酰化的抗人多克隆抗体B A F 213 (R & D Systems) ，或生物酰 化的抗小鼠多克隆抗体B A F 413 (R & D Systems) 二抗稀释缓冲液： 1%(饥 /7) 3 3 八 / 0 . 0 5 % 〇 ^ ) ? 8 3 配 制 的 丁 评 沈 1 1 2 0 〇 / 7 7 ) 辣 根 过 氧 化 物 酶 （H R P ) 结 合 的 链 亲 和 素 （Jackson Immunoresearch) V 底物溶液 检 测 板 :9 6 孔圆底聚乙烯微量板（Dynatech) 9 6 孔聚苯乙烯微量板 微 量 板 洗 涤 仪 （如 E lcatech的 Elcawash) 或塑料洗瓶 微 量 板 读 板 仪 （酶联仪） 1 . 用 P B S 稀释包被抗体至浓度为5Mg/m l 。立即加入至检测板，每 孔 加 50M1，盖上盖 子 ， 37°C 孵 育 2h ，或 4°C 过夜。 不 应 有 其 他 蛋 白 存 在 ，防 止 其 与 抗 体 竞 争 性 结 合 板 上 的 结 合 位 点 。 2 . 每孔加入150jul封闭液以封闭包被好的检测板上多余的蛋白结合位点（包被抗体无 需去除）。 37°C 孵 育 20min。 3 . 在封闭检测板的同时，在 9 6 孔板中用R P M I -1 0 完全培养基对10ng/mlI L -13标准品 进 行 2 倍比梯度稀释。设置好板中所要加50^1体 积 稀 释 物 的 位 置 （如 一 行 的 1 1 个 孔），最后一个孔加培养基作为空白对照。每个稀释度做2 个复孔。用盖子或塑料包 被物完全盖住检测板。 4 . 用 R P M I -I O 完全培养基2 倍比系列稀释待测样品， 50M1体 积 ， 2 个复孔。 5 . 采用微量板洗涤仪或洗瓶用洗涤缓冲液彻底洗涤检测板。一个方向上漂洗2〜3s，然 后 旋 转 180°重复洗涤。用力甩出残留液体，在吸水纸上拍干。 6 . 将稀释好的标准品和待测样品转移到检测板中。盖上盖子，室 温 孵 育 2h 。用力甩出 样品，按照步骤5 洗板。 7 . 用二抗稀释缓冲液稀释生物素偶联抗体至lMg/ml。每 孔 加 50M1，盖上盖子，室温孵 育 45min。用力甩出液体，按 步 骤 5 洗板。 8 . 按照生产商说明书用二抗稀释缓冲液稀释H R P 偶联的链亲和素至最佳浓度。每孔加 5〇 W ，盖上盖子，室温孵育45min。用力甩出结合液，按 步 骤 5 洗板。 9 . 每孔加入100/xl新鲜配制的底物溶液，室温孵育50〜60m i n 直到出现绿色。 1 0 . 以 405n m 为测定波长， 490n m 为参照波长用酶联仪读取最佳密度值。 11. 根 据 IL-13数据绘制标准曲线。确定未知样品中IL-13的浓度。确保未知样品的数 据点落在标准曲线的线性范围内。
