Enzyme activation and inhibition experiments

Summary

Understand the activating and inhibiting effects of enzymatic reactions; learn the methods and principles by which activators and inhibitors affect enzymatic reactions. The activity of an enzyme is often affected by certain substances. Some substances can increase the activity of an enzyme and are called activators; some substances can reduce the activity of an enzyme and are called inhibitors. It is worth noting that activator and inhibitor are not absolute, some substances are activators of an enzyme at low concentrations, while they are inhibitors of the enzyme at high concentrations. For example, NaCl can inhibit the activity of salivary amylase when it reaches 1/3 saturation.

Principle

Enzyme activity is often affected by certain substances, some substances can make the enzyme activity increase, called activator; some substances can make the enzyme activity decrease, called inhibitor. It is worth noting that activator and inhibitor are not absolute, some substances in low concentrations for a certain enzyme activator, while in high concentrations for the enzyme inhibitor. For example, NaCl can inhibit the activity of salivary amylase when it reaches 1/3 saturation.

1. Instrumentation:

(1) Test tubes: 3; (2) Test tube rack; (3) Funnel; (4) Beaker; (5) Graduated pipettes: 1 ml x 4, 5 ml x 1; (6) Thermostatic water bath; (7) 10 ml x 1

IV. Operating Procedure:

Take 3 test tubes and operate according to the following table
