Summary

Tear the epidermis of purple onion or radish leaves, put it on a slide, add a little drop of water and then cover the coverslip, observe the cells in the preparation under the microscope, if there is an intact, rose-purple vesicle containing anthocyanin in one of the cells, then use a strip of filter paper to attract the liquid to be added by successive drops of 0.5M sucrose solution

Experimental Procedure

Tear the epidermis of purple onion or radish leaves, put it on a slide, add a little drop of water and then cover the coverslip, observe the cells in the preparation under the microscope, if there is an intact, rose-purple vesicle containing anthocyanin in the cells, then use a strip of filter paper to attract the liquid to add 0.5 M sucrose solution continuously, so that the cells undergo a strong plasmic wall separation, and then cut off the cells with a sharp razor blade in the plane perpendicular to the longitudinal axis of the cells. The cell was then cut off with a sharp blade on the vertical side of the longitudinal axis of the cell, the dissected but undamaged protoplasm was selected under the microscope, a little more sucrose solution was added, and the coverslip was gently pressed with small forceps to show that the protoplasm was ruptured or detached and the vesicles containing the anthocyanins were freed intact.
