Fecal Test Sedimentation Method
Principle
Suitable for the detection of most helminth eggs and some protozoan encystments. Can be categorized as natural sedimentation, centrifugal sedimentation and mercury-iodoraldehyde centrifugal sedimentation. Move 1. (1) Pick 20-30g of feces with a glass rod and filter it through a 40-60 mesh copper sieve into a conical glass measuring cup full of water and let it stand for 25 minutes;
(2) Pour off the upper layer of fecal liquid, leaving only the sediment, and then filled with water, according to every 20 minutes to change the water once, repeated 3 to 4 times, until the upper layer of liquid clarification;
(3) Finally, pour off the upper liquid and take the precipitate on a slide for microscopic examination. If checking the capsule, the water change interval should be extended to 6 hours to change once
2. Centrifugal precipitation method
(1) Place the precipitate in a centrifuge tube after filtering out the crude residue, and centrifuge for 1~2 minutes at 1500~2000 rpm;
(2) Pour off the upper liquid, then add water and sediment mixing, so repeated centrifugation 3 ~ 4 times, until the upper liquid is clear; (3) Pour off the upper liquid, take the precipitate on a slide microscopic inspection
3. Mercury iodine aldehyde centrifugal precipitation method
(1) Take 1g of feces, add appropriate amount (about 10 ml) of mercury iodine aldehyde solution, mix well;
(2) Filter through 2 layers of skimmed gauze, then add 4 ml of ether and shake well for 2 minutes;
(3) Centrifuge (2000 rpm) for 1-2 minutes, then divide into 4 layers: ether, fecal sludge, mercury-iodoraldehyde and sediment;
(4) Aspirate and discard the upper 3 layers, and take the sediment for microscopic examination.
