Flow cytometric analysis of peripheral blood cells in patients with paroxysmal sleep hemoglobinuria

Summary

This experiment is based on "Color Atlas of Practical Flow Cytometry", edited by Shukui Wang and Zhenying Zhou.

Operation method

Flow cytometric analysis of peripheral blood cells in patients with paroxysmal sleep hemoglobinuria

I. Testing methods

By applying monoclonal antibodies such as CD45, CD61, CD64, etc., the position of platelets, lymphocytes, monocytes, granulocytes, erythrocytes zone III, zone II and zone I cells were set respectively.

1. Negative control tube: CD55/CD59 corresponding isotype control. Take 3 μl of whole blood and add CD45+CD61-FITC/IgG1-PE/CD64-TC and CD45+CD61-FITC/IgG2a-PE/CD64-TC.

2. Positive control tube: 3 μl of normal peripheral blood was taken and CD45+CD61-FITC/CD55-PE/CD64-TC and CD45+CD61-FITC/CD59-PE/CD64-TC were added.

3. Patient test tube: 3 μl of patient peripheral blood was directly stained with CD45+CD61-FITC/CD55-PE/CD64-TC, CD45+CD61-FITC/IgG2a-PE/CD64-TC to determine the expression of CD55 and CD59 in each cell population.
