Principle

In consultation with hospital personnel, provide labeled containers with culture media and arrange to have samples taken from the operating room or pathologist.

1. Provide containers containing collection media, clearly labeled, for delivery to the surgical preparation room or pathology laboratory.

2. be prepared and ready for collection.

3. fill collection containers after surgery or send for collection and deliver promptly with exact time of cut and take.

4. deliver the sample to the culture room, where the sample should be wrapped in three layers (from the inside out: sealed tube, sealed plastic bag with gauze-padded liner labeled with name, address, and phone number, with leak-proof lid, and containing a half-tube of antibiotic-containing culture medium). Although the longer the storage time, the more severe the tissue destruction, the biopsy tissue usually survives at 4°C for at least 24 h and even 3-4 days.

5. Record the data from the samples in a handwritten notebook for transfer to a computerized database or directly into a database.

6. Decontamination

Although most surgical specimens are sterile at the time of sampling, problems may arise during subsequent processing. Epidermal tissues (e.g., skin, melanoma) and gastrointestinal specimens are particularly susceptible to contamination, even if they have been decontaminated by decontamination washing prior to skin biopsy and non-intestinal antibiotics prior to intestinal procedures. A medical microbiologist can be consulted about the flora that may be present in the tissues, and antibiotics can be selected for collection and dissociation of the specimen as needed. If the surgical specimen is large (i.e., 200 mg or larger), a brief soak in 70% ethanol (30 s-1 min) will help minimize surface infection and avoid damage to deeper tissues.
