Summary

Liquid media transfer techniques are used for (1) enrichment and observation of pure species of bacteria (2) characterization of bacterial growth in liquid environments (turbid growth, surface growth, or sedimentation growth).

Principle

Due to the diversity of microbial species and metabolic types, there are many types of culture media used for the cultivation of microorganisms, and their formulas and preparation methods are different. However, the procedures for preparing culture media are generally the same, such as the preparation of vessels, the preparation and dispensing of culture media, the making of cotton plugs, the sterilizing of culture media, the making of slants and plates, and the aseptic checking of the culture media, etc. The basic procedures are generally similar.

Caveat

1. The correct hand position for inoculation of bacteria is to hold the bottom of the plate with bacteria in the left hand and the inoculation ring in the right hand.

2. Remove the inoculation ring and sterilize the mouth of the tube by passing it through a flame.

3. After inoculation, incubate at 37°C in a thermostat.

Common Problems

Results Analysis:


1, turbid growth of Escherichia coli bacterial fluid was uniformly turbid, with a small amount of precipitate at the bottom of the tube.


2, precipitation growth of Streptococcus pyogenes bacterial fluid at the bottom of the tube with precipitation, the bacterial fluid without obvious turbidity.


3、Bacterial membrane growth of Bacillus subtilis bacterial solution to form a membrane on the surface.
