3. Longitudinal section of skeletal muscle tissue
3.1 Low magnification observation:
Skeletal muscle fibers are long and cylindrical, arranged parallel to each other and aggregated in bundles. Due to the long muscle fibers, their ends are often not visible in the specimen. Distinguish the two sides of a muscle fiber border, turn high magnification.
3.2 High-magnification observation: (lower the spotlight appropriately and darken the field of view)
Each muscle fiber on both sides of the darker staining for the myofilm (in fact, not a single myofilm, but also includes the outside of the closely adherent basement membrane). There are many oval or elongated nuclei arranged in longitudinal rows beneath the myofibrils, which are noted to be distinct from the nuclei of the surrounding connective tissue cells. Each myofibril is not obvious, but the myofibrils are arranged parallel to each other along the long axis of the myofibril, and the bright and dark bands of adjacent myofibrils overlap with each other, so that the entire skeletal muscle fiber shows transverse stripes of different shades of coloring. The dark bands are dark red, the light bands are light in color, and there is a thin line in the center of the bands, which is the Z line.

4.2 High magnification observation:
Myofibers are polygonal (due to HE preparation), myofibrils are stained deep red, the nucleus is located under the myofibrils, round or oval, there are many red dotted myofibrils within the myofibrils, and the myofibrils are very lightly stained between the myofibrils for the sarcoplasm.
