Descripción general
Pectinesterase belongs to the pectinase enzyme system and is also known as pectinase, pectin methylesterase, or pectin oxidase. It catalyzes the hydrolysis of methoxyl esters in the long chains of pectin, producing low-molecular-weight substances—pectic acid and methanol—thereby increasing the solubility of pectin in water. This enzyme is widely present in higher plants as well as in bacteria and fungi capable of degrading cell walls. It plays a role in endogenously regulating pectin content on plant cell walls and between cells, and holds significant importance and development potential in the food industry.
Assay Principle: Pectinesterase catalyzes the hydrolysis of pectin molecules to release H⁺, causing a decrease in the pH of the reaction system. The system is maintained at a constant pH of 7.8 using an alkaline solution (with phenolphthalein as an indicator to keep the solution pink). The activity of pectinesterase is determined by measuring the amount of NaOH consumed.
