Sortase A is derived from Staphylococcus aureus and is recombinantly expressed in E. coli. This enzyme is a bacterial transpeptidase that specifically recognizes the LPXTG sequence, cleaves between threonine and glycine, covalently links proteins to the bacterial cell wall, and catalyzes the formation of an amide bond between the carboxyl group of threonine and the amino group of cell wall peptidoglycan. It can achieve specific conjugation of proteins with polypeptides/chemical groups and has no exogenous protease activity.
