Sodium deoxycholate (also known as sodium desoxycholate), with a molecular weight of 414.55 and CAS No. 302-95-4, is a commonly used biological reagent belonging to the ionic detergent class. In aqueous solution, it effectively lyses cell membranes, disrupts cellular structures to release intracellular contents, and solubilizes proteins that are poorly soluble in water. It is widely applied in protein extraction and cell lysis procedures. In addition, sodium deoxycholate serves as a key reagent in the classical bile solubility test: bile or bile salts, possessing surface-active properties, rapidly activate bacterial autolytic enzyme systems, accelerating the autolysis of susceptible bacteria such as Streptococcus pneumoniae. This test is an important biochemical assay in bacterial identification. In the agar plate method, a positive result is indicated by the disappearance of colonies; in the tube method, a positive result is indicated by the clearing of the bile-containing culture, while the control tube remains turbid. This test is commonly used to differentiate S. pneumoniae (positive) from other alpha-hemolytic streptococci such as Streptococcus viridans (negative).
