Overview

Sucrose-PBS Solution refers to PBS solutions containing sucrose at concentrations of 5%, 10%, 20% or 30%. This product is suitable for dehydration of biological samples such as tissues and cells in immunohistochemistry (IHC), immunofluorescence (IF), immunocytochemistry (ICC), flow cytometry and other assays. It can also serve as a dehydrating agent and cryoprotectant for special specimens. Gradient dehydration with Sucrose-PBS Solution effectively prevents ice crystal formation inside samples during frozen sectioning, which would otherwise compromise subsequent sectioning and detection procedures.

PBS, short for Phosphate-Buffered Saline, is mainly composed of NaCl, KCl, Na₂HPO₄ and NaH₂PO₄. Featuring buffering capacity, PBS maintains osmotic balance and preserves the morphological integrity of cells and tissues; it is widely used for cell washing in cell culture, fixation and other experimental workflows. Sucrose exerts a dehydrating effect and is commonly utilized as a dehydrating agent and cryoprotectant.

This product is ready-to-use for convenient application, with no need for preparation or dilution.

Operating Procedures:

1. Collect fresh tissues or perfused tissues, then immerse them in fixative solution at a volume 5–20 times that of the tissue blocks for fixation at 4°C for 8–24 hours (duration adjusted according to tissue block size).

2. Transfer tissues to 5% Sucrose-PBS Solution (5–20 times the tissue volume) for dehydration at 4°C over 12–24 hours, with incubation time determined by tissue dimensions. Once tissue blocks sink completely to the bottom of the container, replace the solution with the next gradient dehydrating agent: sequentially conduct dehydration in 10%, 20% and 30% Sucrose-PBS Solution under identical incubation conditions as the 5% formulation. If sectioning is not performed immediately after full dehydration, tissues may be stored long-term in 30% Sucrose-PBS Solution in a refrigerator at 4°C.

3. Proceed with embedding and other downstream procedures as required by the experimental protocol.

Precautions:

1. Sucrose-PBS Solution has been filter-sterilized. Avoid bacterial contamination during handling.

2. Fully fix tissue blocks with immunostaining fixative prior to dehydration. Trim large tissues into small pieces (1.0 × 1.0 × 0.2 cm) before fixation.

3. Immerse tissue blocks in dehydrating agent at a volume ratio of 5–20:1 relative to the tissue to ensure full coverage and uniform dehydration.

4. Dehydration duration varies with tissue size; complete dehydration is confirmed when tissues sink rapidly to the bottom of the solution.

5. For specimens stored long-term in 30% Sucrose-PBS Solution, regularly inspect tissue conditions and complete subsequent processing promptly to prevent mildew and tissue deterioration.

6. This product is restricted to scientific research by professional personnel only. It shall not be used for clinical diagnosis or treatment, food or pharmaceutical manufacturing, nor stored in residential premises.

7. Wear a lab coat and disposable nitrile gloves throughout the operation for personal safety and health protection.
