Tryptic Soy Agar (TSA) used for the enumeration of aerobic bacteria and the detection of settling microorganisms in pharmaceuticals and biological products.
Principle and Purpose: Tryptic digest of casein and papain digest of soybean meal supply nitrogen sources, vitamins, growth factors and other nutrients. Sodium chloride maintains balanced osmotic pressure. Agar acts as the solidifying agent.
Preparation Method: Weigh 40.0 g of this product and add to 1 L of purified water. Heat and stir until completely dissolved. Sterilize by autoclaving at 121 °C for 15 min. Cool to 50–55 °C and pour into petri dishes for later use.
Scope of Application:
| Application | Category |
| Non-selective count | General-purpose |
Formula (g/L):
| Ingredient | Content |
| Tryptic digest of casein (Tryptone) | 15.0 g |
| Papaic digest of soybean meal | 5.0 g |
| Sodium chloride | 5.0 g |
| Agar | 15.0 g |
| Final pH (at 25°C) | 7.3 ± 0.2 |
Microbiological Testing:
Test in accordance with the Chinese Pharmacopoeia: Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus subtilis, Candida albicans and Aspergillus niger.
