BDNF is synthesized as a precursor form (pro-BDNF) and is converted into its active form through proteolytic processing. The precursor protein is first cleaved to pro-BDNF (~35 kDa) by removing a signal peptide, and then to a mature form of BDNF (~15 kDa). Additionally, pro-BDNF can form dimers that run between 50 -60 kDa, and a minor truncated form of pro-BDNF running at ~28 kDa has also been reported (PMID: 11152678, 20630543).
