I. Experimental reagents:
Liquid A: 5% carbolic acid: 10ml
Tannic acid: 2g
Saturated potassium aluminum sulfate solution: 10ml
Liquid B: Alcohol saturated solution of crystal violet
Application solution: 10 parts of liquid A, 1 part of liquid B, mix and store at room temperature.
II.Dyeing method:
1. Treatment of slides: new slides are required, soaked in 95% alcohol for more than 24 hours before use, removed from the alcohol when used, dried with clean gauze and then used.
2. Put two drops of distilled water on the slide, and two smears can be made on one slide at the same time.
3. Take a small colony from the blood plate with an inoculating pick and dot the bacteria on the top of the distilled water on the slide. Generally only a little bit, only allow a very small amount of bacteria into the droplet, do not stir, so as to avoid the bacterial hairs fall off.
4. Leave the slide at room temperature to dry naturally.
5. Add drops of staining solution to the slide and stain.
6. After about 10-15 minutes, slowly rinse off the staining solution with distilled water, when rinsing, should avoid making the metallic luster film on the surface of the staining solution stay on the slide, which may affect the microscopic examination.
7. After the natural drying of the slide, the microscopic examination should start from the edge of the smear and gradually move to the center, looking for the field of view with fewer bacteria, the flagellum is easy to observe, and where the bacteria are dense, the flagellum is blocked by the bacterial body and is not easy to observe.
