Prothrombin time (TT) measurement test

Summary

The plasma to be tested is added to the appropriate amount of thrombin solution, the fibrinogen is converted to insoluble fibrin to determine the time required for coagulation, i.e., the plasma to be tested prothrombin time (TT). This test is a screening test to check the ability of subjects to convert plasma fibrinogen to fibrin. Principle

The plasma to be tested is added to the appropriate amount of thrombin solution, the fibrinogen is converted to insoluble fibrin to determine the time required for coagulation, i.e., the plasma to be tested prothrombin time (TT). The test is to check the ability of the subject plasma fibrinogen into fibrin screening test.

I. Experimental Methods:

1, reagent reconstruction and preservation

Each bottle of TT reagent was dissolved by adding 2.5 ml of distilled water with gentle shaking to obtain about 5 µl/ml thrombin solution.

Experimental note: reagents dissolved 2-8 ℃ storage for 7 days, 4 hours within -20 ℃ frozen storage, can be stable for 30 days, re-soluble please at 37 ℃ quickly melt.

2、Manual Determination

Take 0.2ml of plasma (reference plasma) pre-warmed at 37℃ and add 0.2ml of pre-warmed thrombin solution at 37℃, mix well. Start the stopwatch immediately and record the coagulation time.

3、Hemagglutination Instrument Determination

Normal value: 10-16 seconds, more than 3 seconds is abnormal.
