Overview
Trypsin selectively hydrolyzes peptide bonds formed by the carboxyl groups of lysine or arginine in denatured proteins, making it a crucial digestive enzyme. In addition, trypsin is widely used as an adjunctive treatment for conditions such as pyothorax, hemothorax, surgical inflammation, ulcers, and traumatic injuries, particularly in addressing localized edema, hematomas, and abscesses.
Assay Principle: Trypsin catalyzes the hydrolysis of the ester bond in TAME (Nα-benzoyl-L-arginine methyl ester), releasing a free carboxyl group. This group neutralizes sodium hydroxide in the reaction system, causing a decrease in the solution's pH. Using phenol red as an indicator, the change in absorbance at 555 nm is measured, allowing for the rapid determination of trypsin activity. Within a certain range, trypsin activity exhibits a good linear relationship with the decrease in absorbance at 555 nm.
