CAA0225 is an epoxysuccinyl peptide and highly selective rat liver cathepsin L inhibitor (IC|50|= 1.9 nM), while exhibiting little to no effect on rat liver cathepsin B at concentrations higher than 1000-5000 nM,|in vitro|. During autophagy, it has been shown through immunoblotting that CAA0225 (10 µg/ml) effectively inhibits the degradation of LC3-II and GABARAP compared to control, in nutrient-deprived HeLa and Huh-7 cells.
