Animal electrophysiology experiment

Principle

Electroencephalogram (EEG) is a graph of brain bioelectrical activity, consisting of multiple brain waves, that is amplified by a precision electronic instrument using an electroencephalograph to record the spontaneous bioelectrical potentials of the brain.

By placing electrodes in the corresponding areas of the brain (e.g., hippocampus, cortex), the spontaneous and rhythmic electrical activity of brain cells and the bioelectrical activity generated by the brain's nerve cells are recorded, thus detecting whether there is any disorder or damage to the brain.


Appliance

It is mostly used for the detection of organic lesions in the brain, such as epilepsy, cerebrovascular disease, and intracerebral space-occupying lesions.

Materials and Instruments

7~8 week male C57BL/6J mice, isoflurane anesthesia system
Electrodes, dental cement, surgical instruments, brain stereotactic apparatus

Move

The steps for detecting EEG in mice are as follows:


1, before the experiment, debug the anesthesia apparatus, apply isoflurane anesthesia system to anesthetize the experimental animals, and maintain its concentration at 1~1.5%;


2、After the animals were anesthetized, they were fixed in the brain stereotaxic position, and after cutting off the hair on the top of the skull, the skin tissue was cut longitudinally to expose the skull;


3、Referring to Bregma as the "0" point, take the coordinates of x = ± 3 mm, y = -3 mm; x = -2 mm, y = 3 mm three points, after the fixed point, punch holes, install the diameter of 2 mm screws, the top connected to the copper wire, and then use dental cement to fix it, and wait for the animal to be resuscitated after the operation;


4. On the second day, the ADInstruments bioelectric amplifier was connected to record the EEG activities of the mice in different states, and Labchart 4.2 software was applied to record and analyze the activities.

Common Problems

1. experimental animals with good physiological state and nutritional state should be selected for EEG detection, and before the experiment, experimental animals should adapt to the environment in advance to avoid interference caused by the environment;


2. the amount of anesthesia should be controlled before the experiment to avoid the death or awakening of the animals in the process of burying;


3. pay attention to the cleanliness of experimental equipment in the experiment to avoid cross infection.
