Enrichment medium preparation experiment
Principle
Prepare as for Extreme Medium, but at high pressure for 25 min (unless otherwise indicated) and without dilution.
Materials and Instruments

Glass bottles Flasks
(1) H medium, per liter
10 g tryptone
8 g NaCl
(2) λ broth medium, per liter
10 g tryptone
2.5 g NaCl
(3) NZC broth medium, per liter
10 g NZ Amine A (NZ Amine A)
5 g NaCl
2 g MgCl2 - 6H20
High pressure for 30 min
5 ml 20% casein hydrolysate (casamino acid)
(4) LB medium, per liter
10 g tryptone
5 g yeast extract
5 g NaCl
1 ml 1 mol/L NaOH
(5) Super broth medium, per liter
32 g tryptone
20 g yeast extract
5 g NaCl
5 ml 1 mol/L NaOH
(6) TB (super broth medium), per liter
12 g Bacto Tryptone
24 g Bacto yeast extract
4 ml glycerol
Add water to 900 ml and autoclave, add 100 ml of sterile 0. 17 mol/L KH2PO4 and 0. 72 mol/L K2HPO4.
(7) Tryptic peptone broth medium per liter.
10 g tryptone
5 g NaCl
(8) 2 X TY medium per liter
15 g tryptone
10 g yeast extract
5 g NaCl
(9) TYGPN medium, per liter
20 g tryptone
10 g yeast extract
10 ml 80% glycerol
5 g Na2HPO4
10 g KNO3 2. Pour into a glass vial, loosen the cap and autoclave at 15 lb/in2 for 25 min.
3. Allow the medium to cool to below 50°C (at this temperature the bottle may feel hot when held in the hand but can be gripped), add the other supplements and antibiotics, cool the glass bottle to below 40°C and close the lid tightly.
4. May be stored indefinitely at room temperature.
