Experimental observation of the components of the lymphatic circulation and their physiological/pathological morphology

Summary

This experimental method was obtained from the official website of Chongqing Medical University

Operation method

Experimental observation of the components of the lymphatic circulation and their physiological/pathological morphology

Principle

Grasp the internal structure of the lymphatic system, analyze the lymphatic vessels in the itinerary connected with the lymph nodes, when the body is attacked by viruses, bacteria and other microorganisms, the lymph nodes not only have the function of lymphatic filtration, but also with the other systems of the body, to resist the microbial infections, constituting an important defense device of the human body; in the proliferation and metastasis of malignant tumors and the role of the lymphatic system has an important significance in the killing; The lymphatic system is often attacked during parasitic infections (e.g., filarial filaria), resulting in serious lesions such as "elephantiasis".

Materials and Instruments

Rats
Chloral hydrate, blue dye.
Rat dissection table Scalpel Scissors Tweezers Syringe Cotton swabs

Move

1. Animal weighing and anesthesia: Chloral hydrate (0.36ml/kg) was injected intraperitoneally.

Make rat mesenteric pavements and observe the rat mesenteric lymphatic vessels. Pay attention to observe its valve and movement.

Pigment injection was made between the toe webbing of rats, and the lymph nodes shown were dissected and observed in the groin for morphological tracer observation of the lymphatic vessels.

2. Lymphatic drainage of the lower limbs

(1) 100ul Patent blue was injected subcutaneously between the 1st and 2nd toe webbing, and the skin was incised longitudinally along the dorsum of the foot and the medial side of the calf to the inguinal area after 5 minutes to search for blue-stained lymphatic vessels and lymph nodes. The superficial lymph node ducts of the lower limbs were located in the superficial fascia, mainly accompanying the small saphenous vein and the great saphenous vein.

(2) Trace the lymphatic vessels accompanying the lesser saphenous vein and their injection into the popliteal lymph nodes. The popliteal lymph node is dissected out in the popliteal fossa.

(3) Trace the lymphatic vessels accompanying the saphenous vein and their injection into the inguinal lymph nodes.

(4) Below the inguinal ligament, dissect the inguinal lymph node.

3. Lymphatic drainage of the stomach

(1) Open the anterior abdominal wall and expose the abdominal organs.

(2) Inject (10ul) of Patent blue into the subplasma membrane of the stomach wall near the greater curvature, observe the lymphatic vessels of the stomach wall, and finally inject into the lymph nodes of the abdominal cavity.

4. Lymphatic drainage of the small intestine

Pull out a section of small intestine tube, inject (10ul) Patent blue under the plasma membrane, observe the lymphatic vessels in the mesentery of small intestine, and finally inject into the superior mesenteric lymph node.

5. Tracing the lymph nodes stained in the retroperitoneal space

In the retroperitoneal space, blue-stained abdominal lymph nodes, mesenteric lymph nodes, common iliac lymph nodes, and lumbar lymph nodes were traced.
