A. Mice are routinely anesthetized.
B. The skin from the head and neck to the back of the mice is scraped with a razor blade to remove the hair, and then an appropriate amount of depilatory cream is applied, and after a few moments of waiting, the excess hair is once again removed cleanly.
C. The depilatory agent is removed by wiping with 75% ethanol and the entire depilated surface is cleaned and disinfected.
D. The skin of the back of the mice is gently lifted up by one operator and the skin is not tautly pulled; another operator blocks the sides of the perforator with stiff pieces of paper. The other operator blocks the sides of the perforator with a piece of cardboard and punches two identical holes at a time on the exposed skin of the lifted mouse.
E. Generally, for statistical analysis, two rows of 4 holes can be punched in parallel on the back of one mouse. Successful holes should be smooth around the perimeter, undercircular, and the area of the 4 holes should be similar. The diameter of each hole is measured with a ruler and recorded.
