Principle

Sulfosalicylic acid is an alkaloidal reagent that binds the cation of protein amino groups on acidic sulfonate ions to an insoluble protein salt precipitate

Materials and Instruments

Sulfosalicylic acid aqueous solution

Move

Experimental reagents: 1, sulfosalicylic acid aqueous solution, sulfosalicylic acid 20g, add water to make up to 100ml

Experimental method: Take a test tube and add 2-3ml of clarified urine, add 0.1ml of sulfosalicylic acid reagent and shake gently for 1 minute to observe the results.

Experimental results judgment: negative (a) clear and transparent

Trace (S) black background only see a slight turbidity

Weakly positive (+) Clear white haze turbidity, no particles

Positive (++) Clear turbidity with particles present

Strongly positive (10-10) White haze with indistinct turbidity and small clumps

Strongest positive (++++) Severe turbidity with numerous clots

Caveat

1. The method is more sensitive (0.05-0.lg/L) mucin, B-J protein and other positive reactions2. some drugs such as high-dose penicillin, hypoglycemic drugs false positive3. high concentration of urate in the urine can be false positive4. Strong alkaline urine can be false negative
