Overview
Fumarase, also known as fumarate hydratase (EC 4.2.1.2), is a key enzyme in the citric acid cycle when located in the mitochondria, while the cytosolic form is closely associated with the metabolism of amino acids and fumarate esters. In humans, deficiency of this enzyme can lead to serious health issues, such as fetal brain malformations and hypotonia.
Fumarase catalyzes the conversion of fumarate to L-malate. Under the action of malate dehydrogenase, L-malate simultaneously reduces NAD⁺ to NADH. The activity of fumarase is determined by measuring the rate of increase of NADH at 340 nm.
