Phenol fuchsin staining solution, also known as carbolic acid-fuchsin stain, is primarily used in biology as a stain for observing chromosomal morphology during cell division. As a basic dye, it is readily soluble in water, alcohol, and acetic acid.
Modified Phenol Fuchsin Staining Solution, also referred to as Carbol fuchsin staining solution, shares similarities with acetocarmine stain. Acetocarmine is a commonly used basic dye often employed for nuclear staining, as well as for the fixation and staining of chromosomes. The staining effect of Modified Phenol Fuchsin Staining Solution on Drosophila salivary gland chromosomes is identical to that of acetocarmine. However, using Modified Phenol Fuchsin Staining Solution can improve efficiency and offers the advantages of simplicity and cost-effectiveness. While acetocarmine provides good staining results for chromosomes, it requires a longer staining time.
Modified Phenol Fuchsin Staining Solution is commonly used for squash and smear methods in plant tissue studies. This solution combines the convenience of acetocarmine staining with the specificity of Schiff's reagent, which stains only nuclei and chromosomes. It offers stable and reliable staining results, suitable for chromosomes of various sizes in both plants and animals, including somatic and meiotic chromosomes. The staining is durable and well-preserved. This product is intended for research and experimental use only and must not be used for other purposes.

Procedure (For Reference Only)
Pollen or similar particles: Place a small amount of pollen material on a glass slide, add an appropriate amount of Modified Phenol Fuchsin Staining Solution, and mix thoroughly. Immediately cover with a cover slip and stain for 5–15 minutes. Absorb excess liquid and observe under a microscope.
Animal or plant tissue sections: After routine processing (e.g., deparaffinization to water), apply Modified Phenol Fuchsin Staining Solution for 5–10 minutes. Rinse with distilled water, mount the slide, and observe under a microscope.
Certain fresh plant tissues (e.g., onion epidermis): Directly immerse in Modified Phenol Fuchsin Staining Solution for 5–10 minutes. Rinse with distilled water for 2–3 minutes, place on a glass slide, cover with a cover slip, and observe under a microscope.
