Monensin is a protein transport inhibitor commonly used to enhance intracellular cytokine staining signals by blocking transport pathways during cell activation. It is particularly suitable for intracellular staining of cytokines, as it allows most cytokines to accumulate in the Golgi apparatus/endoplasmic reticulum.
The optimal usage conditions vary depending on the cell type and treatment duration. Typically, protein transport inhibitors should be added to the culture system 4 to 24 hours before the end of in vitro cell activation culture.
Monensin solution is provided as a 1000× concentrated stock and should be diluted to a 1× working concentration in cell culture medium. It is recommended that cells be co-cultured with monensin for no more than 24 hours, as prolonged exposure may lead to cytotoxicity and affect cell viability.
