Overview

Phosphoglucomutase (PGM) plays a key role in carbohydrate metabolism and is ubiquitously present in all living organisms. PGM catalyzes the reversible interconversion of glucose‑1‑phosphate (G1P) and glucose‑6‑phosphate (G6P). During glycogen breakdown, PGM converts G1P to G6P, which then enters glycolysis to produce ATP, or enters the pentose phosphate pathway to generate ribose and NADPH. When cells require glycogen synthesis, PGM converts G6P to G1P. PGM deficiency can lead to disorders related to glucose storage. This kit provides a simple, sensitive, and rapid method for PGM determination. PGM catalyzes the isomerization of G1P to G6P. G6P is then oxidized by glucose‑6‑phosphate dehydrogenase (G6PDH) to generate NADPH, which reacts with a specific chromogenic probe to form a colored product. By measuring the rate of formation of this product at 450 nm, the PGM activity can be calculated.
